چکیده

In the preceding paper (1) the isolation and culture of Chlorella WEB-17 was described. Evidence was presented that this mutant produced three dicarboxylic porphyrins in relatively large amounts. In this paper we report the isolation from the porphyrin mixture of one of these dicarboxylic porphyrins and its characterization as hematoporphyrin isomer IX (Fig. 1). Hematoporphyrin has not previously been reported as a naturally occurring porphyrin. Although traces of porphyrins of low HCl number arise when tissues rich in hemin have been incubated under sterile or non-sterile conditions for prolonged periods of time (2, 3), no evidence supports the statements that hematoporphyrin is one of these pigments. Fe hematoporphyrin or hematoporphyrin has been shown to arise when the prosthetic group of cytochrome c is split off the protein by certain treatments, the thio ether linkages of porphyrin c side chains being replaced by OH groups (4-6).

Mg-protoporphyrin IX and heme control HEMA, the gene encoding the first specific step of tetrapyrrole biosynthesis, in Chlamydomonas reinhardtii.

HEMA encodes glutamyl-tRNA reductase (GluTR), which catalyzes the first step specific for tetrapyrrole biosynthesis in plants, archaea, and most eubacteria. In higher plants, GluTR is feedback inhibited by heme and intermediates of chlorophyll biosynthesis. It plays a key role in controlling flux through the tetrapyrrole biosynthetic pathway. This enzyme, which in Chlamydomonas reinhardtii is e...

Effect of crude oil contamination on biomass and chlorophyll biosynthetic pathway pigments and elements content of Avicennia marina seedling

Mangrove ecosystems of Iran, located in the shores of the Persian Gulf, near one of the busiest oil shipping routes, are exposed to petroleum pollution. To assess the effects of petroleum contamination of sediments on morphophysiological characteristics of mangrove (Avecennia marina) seedlings, a randomized block factorial experiment was conducted with three replicates. Biomass and pigment cont...

N-Methyl Mesoporphyrin IX Inhibits Phycocyanin, but Not Chlorophyll Synthesis in Cyanidium caldarium.

The ability of N-methyl mesoporphyrin IX (NMMP) to block heme synthesis by specifically inhibiting enzymic iron insertion into protoporphyrin IX was exploited to test whether heme is a precursor of the bilin chromophore of phycocyanin (PC). A strain of the unicellular rhodophyte Cyanidium caldarium which forms normal amounts of both chlorophyll (Chl) and PC in the dark was employed to avoid pho...

Present studies with heme-requiring strain 23 showed that prot...

GENETIC CONTROL OF CHLOROPHYLL BIOSYNTHESIS IN CHLAMYDOMONAS Analysis of Mutants at Two Loci Mediating the Conversion of Protoporphyrin-IX to Magnesium Protoporphyrin

In this report we describe two nonallelic Mendelian protoporphyrin accumulating mutants brs-I and bre-1. Results of experiments with these mutants lead us to postulate that porphyrin biosynthesis branches into light and dark steps between protoporphyrin-IX and magnesium protoporphyrin. We hypothesize that the br e locus controls a dark step while the br s locus either controls a step in the mai...
